Genetic Predictors of Ibrutinib-Related Cardiovascular Side Effects in Patients with Chronic Lymphocytic Leukemia
CONCLUSIONS: Our findings suggest possible genetic determinants of ibrutinib-related CVSEs in CLL. If replicated in a larger study, pre-treatment pharmacogenetic testing for GATA4 and KCNQ1 polymorphisms may be a useful clinical tool for personalizing treatment selection for CLL and/or instituting early risk-mitigation strategies.PMID:37738027 | DOI:10.1158/1078-0432.CCR-23-0421

Therapeutic benefits of prophetic medicine remedies in treating hematological diseases (A review article)
Am J Blood Res. 2023 Aug 15;13(4):130-142. eCollection 2023.ABSTRACTHematological disorders are common medical ailments constituting an important cause of morbidity and mortality worldwide, which may be managed efficiently using different prophetic medicine remedies as adjuvants to current therapeutics. Prophetic medicine includes the body of knowledge about medicine that has been derived from the deeds, customs (sunnah), ahadith (sayings), actions, and agreements of Prophet Muhammad, peace be upon him. This review article aims at exploring the magnitude of therapeutic benefits of prophetic medicine remedies as adjuvant tr...

Pharmacological inhibition of the MLL1-menin-interaction aggravates acute kidney injury induced by folic acid and ischemia/reperfusion in mice
In this study, we investigated the role and mechanisms of MLL1 in murine models of acute kidney injury (AKI) induced by folic acid (FA) and ischemia/reperfusion (I/R). Injury to the kidney elevated expression of MLL1, Menin, WDR5 and H3K4Me3, which was accompanied by increased serum creatinine (Scr) and blood urea nitrogen (BUN), renal tubular injury and apoptosis. Pharmacological inhibition of MLL1 activity with MI503 to disrupt the interaction between MLL1 with Menin further increased Scr and BUN levels, enhanced expression of neutrophil gelatinase associated lipocalin and kidney injury molecule-1, and induced more apopt...

Design and synthesis of novel 1,2,3,4-tetrazines as new anti-leukemia cancer agents
Chem Biol Drug Des. 2023 Sep 20. doi: 10.1111/cbdd.14328. Online ahead of print.ABSTRACTA series of novel 1,2,3,4-tetrazines were designed and synthesized. 1 H-NMR spectroscopy, 13 C NMR spectroscopy, and HRMS were used to determine the structures of this novel compounds. Computational approaches suggested that DHFR is a putative target for the newly synthesized 11 compounds. Extensive molecular dynamics simulations followed by molecular docking simulations were employed to evaluate DHFR as a potential target protein. The anticancer activities of the compounds were evaluated against five different types of leukemia cell li...

Immune checkpoint molecule DNAM-1/CD112 axis is a novel target for NK-cell therapy in acute myeloid leukemia
In conclusion, the modification of the DNAM-1/CD112 axis in NK cells may be an effective novel immunotherapy for AML. Furthermore, these results suggest the potential of the expression levels of these molecules as prognostic markers in AML.PMID:37731380 | DOI:10.3324/haematol.2023.282915

Exosomal circKIAA1797 Regulates Cell Progression and Glycolysis by Targeting miR-4429/PBX3 Pathway in Gastric Cancer
Biochem Genet. 2023 Sep 20. doi: 10.1007/s10528-023-10529-z. Online ahead of print.ABSTRACTIn recent years, circular RNAs (circRNAs) are extensively studied in the progression of various types of cancer, while the mechanism of circKIAA1797 is rarely studied in gastric cancer (GC). Hence, this research aimed to investigate the expression of exosomal circKIAA1797 and its biological function in GC cells. Exosomes were extracted from the serum of GC patients and identified by transmission electron microscopy (TEM) and nanoparticle tracking analyzer (NTA). CD81, CD63, Bcl-2, Bax, and pre-leukemia transcription factor 3 (PBX3) p...

Single-cell genotypic and phenotypic analysis of measurable residual disease in acute myeloid leukemia
Sci Adv. 2023 Sep 22;9(38):eadg0488. doi: 10.1126/sciadv.adg0488. Epub 2023 Sep 20.ABSTRACTMeasurable residual disease (MRD), defined as the population of cancer cells that persist following therapy, serves as the critical reservoir for disease relapse in acute myeloid leukemia and other malignancies. Understanding the biology enabling MRD clones to resist therapy is necessary to guide the development of more effective curative treatments. Discriminating between residual leukemic clones, preleukemic clones, and normal precursors remains a challenge with current MRD tools. Here, we developed a single-cell MRD (scMRD) assay ...
